L-Theanine Benefits: What the Research Shows
L-theanine is an amino acid found almost exclusively in tea leaves. It crosses the blood-brain barrier, raises alpha-wave activity in the brain, and shifts the balance of several neurotransmitters, all without causing drowsiness. That combination of calm and clarity is why researchers keep studying it and why it appears in a growing number of focus-oriented nootropic supplements.

What is L-theanine?
L-theanine is a non-protein amino acid. The tea plant (Camellia sinensis) produces it, and a few mushroom species contain it, but it does not appear in any common food outside tea. Green tea carries more than black tea because the fermentation process breaks some down.
Structurally, L-theanine resembles glutamate, the brain's main excitatory neurotransmitter. That similarity lets it interact with glutamate receptors, but instead of exciting neurons it tends to moderate their activity. The body does not make L-theanine on its own, so the only way to get it is through tea or a supplement.
How does L-theanine work in the brain?
L-theanine changes brain chemistry through at least three routes.
First, it increases alpha-wave activity. In an EEG study by Nobre and colleagues, participants who took L-theanine showed a significant rise in alpha-band oscillations compared to placebo. Alpha waves are the electrical signature of relaxed attention: present when you are awake and focused, absent when you are asleep or anxious. This is the mechanism behind the "calm but alert" feeling that tea drinkers describe and that coffee alone does not produce.
Second, L-theanine influences several neurotransmitters. It raises GABA, serotonin and dopamine levels in the brain. GABA is the main inhibitory transmitter and tends to reduce anxiety. Serotonin and dopamine are involved in mood and reward. The net result is a shift toward calm focus rather than sedation or excitement.
Third, L-theanine partially blocks glutamate receptors, which moderates the excitatory signals that contribute to stress and overstimulation. A 2007 study by Kimura and colleagues proposed that this inhibition of cortical neuron excitation is the mechanism behind the reduced stress responses they measured in their trial.
What are the proven benefits of L-theanine?
The benefits below have human trial evidence. Where a finding rests on a single small study, the text says so.
Focus and attention
L-theanine improves attention, and the effect is strongest when L-theanine is paired with caffeine. In a randomised trial by Owen and colleagues, 100 mg of L-theanine with 50 mg of caffeine improved both speed and accuracy on an attention-switching task at 60 minutes, and reduced distraction in a memory task at both 60 and 90 minutes. Caffeine alone improved only alertness and accuracy, with no speed gain. The combination outperformed caffeine on every attention measure.
L-theanine alone also shows attention effects. A 2021 RCT by Baba and colleagues tested L-theanine in middle-aged and older adults and found that a single dose shortened reaction time on the Stroop test and improved correct answers on a working memory task. The authors concluded that "L-theanine may contribute to improving attention, thus enhancing working memory and executive functions."

Stress and anxiety reduction
L-theanine reduces subjective stress and anxiety in controlled settings. In the Hidese 2019 RCT, 30 healthy adults took 200 mg of L-theanine daily for four weeks. Trait anxiety scores dropped significantly compared to placebo (p = 0.006), and depression scale scores also fell (p = 0.019). These were not patients with diagnosed anxiety, but healthy volunteers reporting everyday stress.
The physiological side matches. Kimura and colleagues gave participants L-theanine before a mental arithmetic task designed to induce stress. Heart rate rose less, and salivary immunoglobulin A, a marker of acute stress, dropped compared to placebo. The effect appeared to come from reduced sympathetic nervous system activation.
Sleep quality
L-theanine is not a sedative, and the trials are clear on this point. It does not shorten the time it takes to fall asleep. What it does is improve the quality of sleep once you get there.
In the Hidese 2019 trial, Pittsburgh Sleep Quality Index scores improved during the L-theanine period (p = 0.013), with better subscores for sleep disturbance and less use of sleep medication. A larger trial in boys with ADHD (Lyon et al. 2011, n = 98) found that 400 mg of L-theanine per day raised sleep percentage and sleep efficiency, while sleep latency stayed the same. The authors described L-theanine as "safe and effective in improving some aspects of sleep quality."
The alpha wave mechanism may explain why. Alpha activity is part of the transition from waking to sleep, and people who produce more alpha activity during this window tend to report better sleep quality. L-theanine appears to make this transition smoother rather than forcing it.
Blood pressure under stress
L-theanine reduces the blood pressure spike that some people experience during mental tasks. In a crossover trial by Yoto and colleagues, 200 mg of L-theanine lowered both systolic and diastolic pressure during a stressful mental task, but only in participants who were high-stress responders to begin with. It had no effect during a physical stress test (cold water on the hand) and no effect in people whose blood pressure stayed stable under stress.
This is a narrow, real finding: L-theanine takes the edge off the cardiovascular stress response in people who react strongly to mental pressure. It is not a treatment for hypertension.
Cognitive function in healthy adults
Beyond attention, L-theanine may support broader cognitive performance. In the Hidese 2019 trial, four weeks of 200 mg daily improved verbal fluency scores (p = 0.001) and executive function scores (p = 0.031). The improvements were most pronounced in participants who started with lower baseline scores.
This is one trial with 30 participants, so treat it as a signal rather than a settled finding. A larger study with the same design would either confirm or narrow the effect.
How much L-theanine should you take?
Most studies use 100 to 200 mg for cognitive effects and 200 to 400 mg for stress and sleep. The Lyon sleep trial used 400 mg daily. The Hidese stress and cognition trial used 200 mg daily. Attention studies with caffeine typically pair 100 mg of L-theanine with 50 to 100 mg of caffeine.
A cup of green tea contains roughly 25 to 60 mg of L-theanine, depending on the type, brewing time and water temperature. To reach the 200 mg dose used in most clinical trials, you would need four to eight cups, which also means a lot of caffeine. Supplements deliver the dose directly.

What are the side effects of L-theanine?
L-theanine has a clean safety record in the published trials. No study has reported serious adverse effects at doses up to 400 mg per day over several weeks. The US Food and Drug Administration granted L-theanine Generally Recognized as Safe status.
A few points to keep in mind:
Blood pressure medications. Because L-theanine can reduce the blood pressure spike during stress, stacking it with blood pressure medication could, in theory, lower pressure too far. No trial has documented this interaction, but it is reasonable to mention it to a doctor.
Stimulant medications. L-theanine modulates some of the same neurotransmitter systems that stimulants act on. If you take a prescription stimulant, check with a prescriber before adding L-theanine.
Pregnancy and breastfeeding. No trial has tested L-theanine supplements in pregnant or breastfeeding women. Tea consumption during pregnancy is generally considered safe in moderate amounts, but supplement doses deliver far more L-theanine than tea, and the research gap means caution is warranted.
Where do you find L-theanine naturally?
Tea is the only common dietary source. Green tea has the highest concentrations because the leaves are less processed. Matcha, which uses the whole ground leaf, delivers more per serving than steeped green tea. Black tea and oolong contain L-theanine too, but fermentation reduces the amount.
A few mushroom species, including Boletus badius, contain L-theanine in small amounts. These are not a practical dietary source.
In supplement form, L-theanine is usually sold as a standalone capsule or as part of a formula that pairs it with caffeine or B vitamins. The L-theanine used in supplements is produced by enzymatic synthesis, and the product is chemically identical to the L-theanine in tea.
